To work within the group of Dr Ingo Greger at the MRC Laboratory of Molecular Biology (LMB), within a programme aimed at investigating AMPA glutamate receptor (AMPAR) operation and organisation at CA1 synapses. Specifically, to investigate the nano-positioning of AMPARs and their repositioning during LTP (long-term potentiation), using a combination of synaptic recordings (Penn et al., Neuron 2012; Watson et al., Elife 2017; Ho et al., J. Neurosci. Meth. 2020) and super-resolution imaging of hippocampal synapses (Watson et al., Nature Comms 2021). The successful candidate will work in close collaboration with a PhD student and the LMB Light-microscopy team led by Dr. Nick Barry. We also closely collaborate with Cambridge University on studies of hippocampal synaptic plasticity mechanisms. In this project, we will characterise signalling and nano-positioning of synaptic/ dendritic glutamate receptors under baseline conditions and in response to LTP. LTP will be induced using various protocols including 2-photon glutamate uncaging onto individual dendritic spines. Towards this wider aim, we have established various state of the art approaches including 3D-STORM, STED and CRISPR/ Cas-9 genome editing. The successful candidate will have their own electrophysiology rig, and will have an opportunity to set-up a 2-photon rig (currently being purchased). To work within the group of Dr Ingo Greger at the MRC Laboratory of Molecular Biology (LMB), within a programme aimed at investigating AMPA glutamate receptor (AMPAR) operation and organisation at CA1 synapses. Specifically, to investigate the nano-positioning of AMPARs and their repositioning during LTP (long-term potentiation), using a combination of synaptic recordings (Penn et al., Neuron 2012; Watson et al., Elife 2017; Ho et al., J. Neurosci. Meth. 2020) and super-resolution imaging of hippocampal synapses (Watson et al., Nature Comms 2021). The successful candidate will work in close collaboration with a PhD student and the LMB Light-microscopy team led by Dr. Nick Barry. We also closely collaborate with Cambridge University on studies of hippocampal synaptic plasticity mechanisms. In this project, we will characterise signalling and nano-positioning of synaptic/ dendritic glutamate receptors under baseline conditions and in response to LTP. LTP will be induced using various protocols including 2-photon glutamate uncaging onto individual dendritic spines. Towards this wider aim, we have established various state of the art approaches including 3D-STORM, STED and CRISPR/ Cas-9 genome editing. The Centre’s goal is to facilitate and provide research excellence through a focus on world-class facility capabilities matched with wide array of staff expertise in the life and material sciences. CAM is a purpose-built facility which comprises 8 research professional and administrative staff supporting a diverse range of state-of-the-art instrument platforms and techniques in basic and advanced light, electron and X-ray microscopy and analysis The Life Science instrument suite ranges from live cell imaging, high-throughput slide scanning and two confocals (ZEISS LSM780 and ZEISS LSM800/Airyscan) on the light microscopy (LM) side, two newly acquired 200kV Life Science TEMs (JEOL F200 and JEOL cryoARM200) to several high resolution FESEMs including a recently installed ZEISS Crossbeam 550 (FIB-SEM). We also provide and support a correlative approach of LM and EM for both room and cryo temperature research projects. The cryo suite is supported by a host of available cryo sample preparation instrumentation and procedures, which includes a Leica EM GP2 plunge freezer, a Leica EM ICE high pressure freezer, a Leica AFS freeze substitution and a Leica UC7 cryo-microtome. These tools, combined with the cryoEM instrumentation enables CAM to provide single particle analysis (SPA), cellular architecture of molecular machines and complex 3D volume imaging of tissue samples (cryo tomography and cryo volume imaging). Exposure to working with nucleic acids (e.g. plasmid DNA, messenger RNA, siRNA, CRISPR components) and proteins. Starting wage for this position is $22.00 + 1.00 shift differential for NIGHTS! The Filtration Operator has the responsibility for running the RO/Separator/UF/Nano/Pasteurizer’s in an efficient and productive manner. Communicate with all departmental operators to efficiently manage silo space and production schedules. Any issues should be reported to a shift lead/supervisor/manager in a timely manner. Must have ability to operate NIR effectively. Must pull liquid samples and use analysis for adjusting operation parameters. Monitor, plan and track all incoming product for upcoming production runs. Check all silo temperatures and solids; document this information on the RO / UF Daily production report. Trace product lines and track the locations of all products being transferred. Maintain the filtration equipment assuring that all product specifications meet specified parameters. Maintain proper documentation for production Runs and CIP’s. Monitor the filtration equipment and silo areas for leaking gaskets, airlines, pump seals and any other operation deficiencies and report immediately to maintenance/shift lead/supervisor/manager. Must operate all CIP3, CIP4 and CIP6 systems within the plant. Work along with BK Dryer Officer assisting in operation of UD and CIP. Maintain clean silos, lines and documentation.
